Relationship to mTOR: Leucine is a well-characterized direct activator of mTORC1 via Sestrin2-GATOR2/GATOR1-Rag signaling (see /rag-gtpases upstream, /s6k1 downstream). Restricting the combined BCAA class reduces this input. Stored as a SEPARATE entity from isoleucine-only restriction (/isoleucine-restriction) and from total protein restriction (/protein-restriction) -- see datamodel warning.

Safety notes

No established human BCAA-restriction longevity dose exists. BCAA restriction has separately been studied in specific clinical-metabolic-disease contexts (e.g. management of maple syrup urine disease), which is a distinct clinical question this longevity-evidence page does not address.
